Evaluating an Interleukin-1β Injection to Induce Degeneration in the Rat Lumbar Intervertebral Disc NSF Summer Undergraduate Fellowship in Sensor Technologies

چکیده
Early signs of degenerate intervertebral discs include a decrease in the glycosaminoglycan (GAG) content within the nucleus pulposus and the annulus fibrosus, which has been linked to an alteration in the mechanics of the disc. However, the mechanisms for this reduction in GAG are currently unknown. An animal model that demonstrates disc degeneration would be useful not only for understanding the process of degeneration, but also for exploring methods of treatment or regeneration. Specifically, this study aimed to develop an in-vivo murine model resembling early intervertebral disc degeneration by introducing Interleukin-1β (IL-1) into the lumbar spine. IL-1 was injected into the nucleus pulposus of the lumbar intervertebral discs of rats, and these discs were evaluated using mechanical testing, biochemical assays, and histology at one and four weeks post-injection. A control group and a sham injection of PBS were used to evaluate the significance of the IL-1 injection. At one week, a reduction was seen in nucleus GAG content in the IL-1 discs. At four weeks, glycosaminoglycan restoration was seen in the IL-1β treated discs, suggesting the possibility of recovery of the disc. TABLE OF CONTENTS

Degeneration of the lumbar intervertebral discs is strongly implicated as a cause of low back pain, and may also lead to impaired mobility. A lack of understanding of the pathomechanisms that underlie degeneration limits our ability to develop biological treatments that both alleviate painful symptoms and restore function. The process of degeneration is characterized by up-regulation of pro-inf...
متن کاملDegenerative Disc Disease: A Review of Cell Technologies and Stem Cell Therapy
Background & Aim: Low back pain is broadly documented as one of the most widespread pathologies in the advanced domain. Although the reasons of low back pain are uncountable, it has been meaningfully related to intervertebral disc degeneration. Present therapies for Intervertebral Disc (IVD) degeneration such as physical therapy and spinal fusion reduce symptoms' severity, but do not treat the ...
متن کاملEffects of Morphine Dependency on Intervertebral Disc in the Rat
Background: A large proportion of patients presenting with lumbar disc disease are opium addicts in our region. It was, therefore, hypothesized that morphine might affect the intervertebral disc. We investigated the histological findings of morphine dependency on intervertebral discs in rat. Methods: Forty NMRI adult male rats (230-250 g) placed on ordinary diet and received aqueous solution of...
متن کاملAssociation of Rs1676486 genetic polymorphism and lumbar disc degeneration in Iranian population
Background: lumbar disc degeneration is a multifactorial degenerative disease which is affected by genetic inheritance and environmental factors. Type XI collagen is important for organization of the extracellular matrix and cartilage collagen construction. Rs1676486 is a SNP that causes the conversion of C-T, resulting in a change in the expression of the collagen 11 alpha chain. The T allele ...
متن کاملMeasuring Dimensions of Lumbar Intervertebral Discs in Normal Subjects
Introduction: Disc degeneration is a natural aging process characterized by changes in appearance and chemical structure of the disc. This degeneration leads to back pain. In regard to the limitation of current therapeutical methods for patients with degenerated discs, arthroplasty has been suggested as an alternative method. To manufacture artificial discs for Iranians, determining normal lumb...
